Human morality Essay
Morality has often been associated with the roles performed by the concept of nature and nurture in relation to the source of moral dispositions. It has been expressed that evolution has provided human beings preconditions of morality, yet it is solely during engagement with an individualââ¬â¢s culture that these preconditions are converted to pure morality. Evolution considers human beings as one of the animal species and thus it is not easy and simple to detach ourselves from the rest of the other biological species in terms of our morals and conducts (Crawford and Salmon, 2004). The concept of morality is thus not an essential part of being human, but its presence makes us adjust to our environments, as well as act in a rational way. Morality also allows human beings to care for other individuals, as well as experience the feeling of wanting something. More importantly, morality provides us an additional characteristicââ¬âto want to be decent individuals. There has been significant debate with regards to the implications of morality and that of the concept of evolution coined by Charles Darwin. It has been described in the history of Social Darwinism and the concept of eugenics that it is possible to choose which genes should further proliferation in the next generations and those genes that confer undesirable traits be left out to go extinct in the human race. This concept of eugenics has been attempted in eradicating future generations of criminals as well as sickly individuals that carry heritable medical disorders. Unfortunately, morality is also influenced by the concept of Social Darwinism, wherein there is huge misinterpretation of the theory of evolution, especially that of the concept of fitness. One major issue of misinterpretation is that the followers of Social Darwinism strongly equate the word good to the condition of being highly evolved. Hence any individuals that are not behaving in a lawful manner such as criminals are immediately classified as individuals that belong to a lower level in the taxonomy of human evolution. This concept should thus be revisited so that the term fitness be only associated with physical fitness of an organism as it evolves through time. It should also be known that the fitness of an organism is strongly influenced by an environment, wherein an organism that lives in a harsh environment learns to survive in cruel ecological conditions, and in the case of human beings, in poor social and environmental conditions. Moral dispositions among human beings evolve through the process of natural selection, hence the results of evolution are simply considered to be that of the selfish trait. It is generally accepted that the human brain is comprised of structures that have evolved to follow that ââ¬Å"either-orâ⬠way of thinking, wherein the mind is put in a blank non-thinking conditions and the rest of the functions of the brain are at an innate condition. There are also some genetic misinterpretations with regards to human nature, wherein there is so much attention on the transmission of intelligence and other superb human capabilities that is known be better understood that there is also a concept of determinism wherein an individual carries this need to be compassionate to other individuals because they feel a certain peace and tranquility in their bodies, including the brain and the heart. Thus there is an interaction between the environmental and the genes of an individual, resulting in the personality of an individual to sacrifice his own benefits for the good of others. The concept of eugenics thus can not be fully applied to human morality because this concept is affected by multiple factors, mainly by the genetic makeup of the individual, as well as the environmental effects on the physiological brainwork of an individual. Google Books Essay
The Google Books project has been a working progress ever since Google was created. The co-founders of Google, Sergey Brin and Larry Page had been working on a research project that was supported by the Stanford digital Library Technologies Project in 1996. Google intends to scan every book ever published and make all of the text searchable so that people can find the relevant information they need about book. They want to make books more accessible to the public and create an easy mechanism of sorting a bookââ¬â¢s content and relevance to a subject. In 2002 a secret ââ¬Å"booksâ⬠project was launched and research was underway to identify the challenges that lay ahead of them. Over this period, Googlers discovered a quick and harm free way to scan books and began to meet with Libraries to begin the digitalization of books. In December 2004 Google announces the launch of the ââ¬Å"Google Printâ⬠Library Project thanks to partnerships from Harvard, The University of Michigan, The New York Public Library, Oxford and Stanford. Together it is said that these libraries exceed 15million volumes. In 2005 Google Print is renamed Google Books which is a more fitting title as it better explains itââ¬â¢s use. With the launch of Google Books and its fast development many will argue of the advantages and disadvantages of the site. The whole project seems a little bit overly ambitious and it obviously has many flaws in its system. It is a timely process to scan hundreds of millions of books and the pivotal question here is ââ¬Å"Are Google books doing it right? â⬠Scanning books is an extremely time consuming process so once Google books have done it, it seems unlikely that the books will be rescanned. If some of the books are not scanned properly, important literature and information could become obscured or lost through the process of digitalization. Geoff Nunberg (2009) published an article Google books: A Metadata Train Wreck and pointed out many errors in the system. One example being that he googled the name of an author and restricted the search to the works published before their year of birth. It was found that 182 hits came up for Charles Dickens alone. The Chief Engineer for Google Books, Dan Clancy claimed that the incorrect dates where the fault of the libraries. However, when the matter was investigated further it shows that the first ten full read books published before 1812 and that mention Charles Dickens are correctly dated in the catalogues that they had come from. Although one can argue that the correct information is given on the title page, there have been some other inexcusable errors too. Google Books has classified many of its books incorrectly and once again Dan Clancy has claimed that both the libraries and publishers where to blame because the classifications were drawn from the BISAC codes that is given to booksellers. BISAC codes have only been around for about 20 years meaning that any book that was put in the wrong category before this time is a mistake of Google themselves. Google have decided to take on an extremely large project but it seems apparent that they are not doing it very well. They are quick to push the blame on others and the whole project is based more towards commercialism rather than to help make knowledge available to the world. Project Gutenberg was one of the first ââ¬Å"digitalâ⬠libraries and was created by volunteers. This project seems to focus more on the importance of literature and the quality of the books available are much greater than those on Google Books. The books are proof read by human beings and their workers are not paid which is a clear sign that they actually care about making books more available to people. Google Books produces books in a much larger mass but they should be aware that people will value ââ¬Å"quality over quantityâ⬠most. Google quickly scan these books and itââ¬â¢s obvious that they rarely check them for errors. In Paul Duguidââ¬â¢s (2007) essay Inheritance and Loss? A Brief Survey of Google Books, He addresses the Google books system hands on by using Laurence Sterneââ¬â¢s The Life and Opinion of Tristram Shandy as an example. He choose the first link that appeared in the search engine and claims his results were as follows; The book he was examining did not start with the word ââ¬Å"wishâ⬠meaning that the left hand side of the page that had the word ââ¬Å"Iâ⬠was missing. On page seventeen the left hand side of the page is not legible because the gutter of the book is blocking the first few letters and by page twenty-seven, Sterne quoted Hamletââ¬â¢s phrase ââ¬Å"alas, poor Yorick! â⬠and inserted a black page of mourning. However the version that is on Google books has left out this page and is somewhat ignorant to the fact of how iconic it is to the astute reader. On further investigation of Duguidââ¬â¢s essay I clicked on the links that were given to the book and realised that it was no longer a link to the book. I then searched Tristram Shandy just had Duguid had done into Google Books. I clicked on the first link which is the same Harvard edition that Duguid was referencing and discovered that the first page had the word ââ¬Å"Iâ⬠before ââ¬Å"wishâ⬠and page seventeen was now fully legible. Although some corrections had been made the black page that was to follow on from page twenty seven has still not been inserted. This is perhaps due to the fact that the people scanning these books are not scholars themselves. It is very easy to recognise a page with a missing word or one that is not fully legible but many would mistake a black page as an error in printing. Another flaw in the digitalization in books is the actual book itself. There is something so pleasant about flicking through a book and holding it in your hand while you read. The book in its own physically is magnificent, depending on how old it is it could have been passed on from generation to generation. The book itself is a story in its own right. Throughout its lifespan the book can acquire various annotations, signatures and other interesting characteristics. There has also been a lot of conflict with regard to the publishing industry and the digitalization of books. Google has offered to provide a search engine what they aspire to be every book ever published but for those which are copyrighted and cannot be viewed online, Google provides the option to purchase them online through sites such as Amazon or Barnes. In January 2007, Google held a conference on the future of the publishing industry. The conference quoted Charles Darwin and projected it on a screen: ââ¬Å"It is not the strongest of the species that survive, nor is it the most intelligent, but the ones most responsive to change. â⬠Toobin (2007) states in an article Googleââ¬â¢s Moon Shot: ââ¬Å"As Laurence Kirschbaum, a long time publishing executive who recently became a literary agent, told me at the conference, ââ¬Å"Google is now the gatekeeper. They are reaching an audience that we as publishers and authors are not reaching. It makes perfect sense to use the specificity of a search engine as a tool for selling books. â⬠â⬠This statement has a lot of truth because since the growth of technology, the popularity of books has fallen drastically. People in the 21st century care more for mindless television shows and tacky magazines than a good well written piece. Reading books challenges the mind and fuels the imagination and by incorporating literature with technology it is a great attempt to try and revive such an excellent thing. Despite Googleââ¬â¢s attempts, it looks as if they are not doing a good job. Many authors and publishers filed a lawsuit against Google Books claiming that Google has violated their copyrights by scanning the books, creating an electronic database and displaying short excerpts without their permission. The Authors Guild filed a lawsuit against Google Books alleging copyright infringement and after four years of discussion a settlement was finally reached in 2009. It was decided that Google was allowed to copy, display and sell millions of books that were out of print but still in copyright. However the agreement was reviewed several times and was summarily rejected in March 2011. This gave organizations a chance to voice their concern about the privacy policy for users of the system. Google claims that it has reviewed its privacy policy and that an advance policy has been created for Google Books but it seemed that the policy still left a large number of gaps and something appeared to be quite vague. On September 4th 2009 the Electronic Privacy Information Centre (EPIC) entered a motion to intervene in the Google settlement case to help the readers of Google Books regarding their privacy online. EPIC states that readers will be required to part with particular information that will be stored in a database to create detailed profiles of preferences of the reading with regard to their purchases and browsing. Marc Rotenberg appeared in court on February 18th 2010 and stated that: ââ¬Å"A person at any library or any university in the United States that attempted to retrieve information from Googleââ¬â¢s digital library would be uniquely tagged and tracked. There is simply no precedent for the creation of such powerâ⬠. The court rejected the settlement but it did however state that Google should review itââ¬â¢s privacy policy to better protect its users. Google should not have the right to disclose any information to government or third parties and secure browsing should be a priority. Many people do not realise that these profiles are being created or that their privacy is being invaded. If this is supposed to be the library of the future, then it should be dealt with in the correct manner. People should be able to browse through books anonymously and have their own thoughts kept private. Google Books seem to have rushed the whole process of scanning such a vast amount of literature and by doing so they seemed to have forgotten about ââ¬Å"quality over quantity. â⬠It seems that the dream of creating a digital library will remain one for the foreseeable future due to the numerous flaws that the system has. Whilst Google Books are trying to correct their many errors it is apparent that the whole project was done quite carelessly and insufficiently. It is evident that Google Books motive leans more to the commercial side of things rather than making knowledge available to a wider audience. This is particularly clear with regards to their privacy policy. Google will suggest recommended pages and sites by analysing all of your browsing. The option to purchase a book that is not available online makes Google Books more of a digital bookstore rather than a digital library. Arguing Both Sides- Gay Marriage
The issue of gay marriage stirs up controversy, which makes it a national problem. Although some states, and many people, now accept gay marriage, gays still face many obstacles. Now that gay marriage Is allowed, the fight to legalize gay adoption grows. Even though the Idea of gay couples has become more accepted across the united States, gays adopting children remains a major controversial Issue. Occurring since American colonial times, adoption provides children for childless couples. Back then, adoptive parents were protected from dealing with infertility and acing the differences between being a parent through adoption versus being a parent by birth. Objections to adoption did not exist during colonial times as long as adoptive parents provided care and support for the child. Therefore, parents who properly care for adopted children were assured no judgment or speculation would rise. The primary objection to gay adoption today is that it goes against nature, which says the primary caregivers for children should be the man and woman who created the child. In cases where a child constantly switches to dfferent foster homes as pposed to a stable home run by a gay Individual or couple, a caring person must choose the latter. The only method of conception for human beings requires Involvement of both a male and a female. Gays and Infertile heterosexual couples turn to child adoption when wanting to start a family. Opposers of gay adoption believe that marriage without the potential to reproduce is a sin. Discriminants think that if you cannot reproduce naturally, you are not meant to have children. People against gays refer to the bible which says a man will be united to his wife. However, God created gays as well and anti-discrimination laws declare that gays and heterosexuals should be treated equally. As stated in the Declaration of Independence, all men are created equal. Adoption means to legally bring someone into your family and raise them like you would your own child. Adopting a child involves the creation of a parent-child relationship between individuals who are not naturally related. A childs best interests are the most Important factors considered during adoption. The type of person allowed to adopt children, however, continues to cause heated debates. In the essay Counterpoint: In Defense of Gay and Lesbian Parents, John Pearson provides support for gay couples wanting to be parents. Pearson is a novelist who was educated at Kings College School. The essay Point: Children Suffer with Same-Sex Couples, written by Lynn-nore Chittom and co-written by Geraldine Wagner, opposes what Pearson supports about gay adoption. Chittom is a self-employed freelance writer. Wagner is a graduate of the State university of New York at Fredonia, teaching sociology and technical and professional writing. Pearson states, A huge number of Americans that might make excellent parents are being enied the legal right to adopt (Pearson, par. 5). There are many different types of families In the United States. Some examples are as follows: step families, single parent families, and adoptive families and so on. Therefore, Pearson wonders why heterosexual families and Individuals should be thought to be able to provide better care and surroundings for a child than a gay or lesbian couple. Nearly 520,000 children await adoption in foster care, but each year only 50,000 find permanent adoption by gays could potentially raise the number of children who find a home. When making a decision on adoption, Pearson believes aspects like a familys lifestyle and ability to care for a child should be more important than the sexual preference of who wants to adopt the child (Pearson, par. ). On the flip side of the argument, Chittom talks about how children are best reared in environments with both a mother and a father, referring to the Fifth Amendment which clearly states Honor your father and your mother. (Chittom, par. 4). However, many gay couples form more stable relationships than heterosexual couples. Pearson opposes Chittom with he idea gays may be even better at child-rearing because gays are n ot raising a child conceived on accident, but rather one fought to obtain. Opposers of gay marriage strongly believe gay partners cannot care for a child as well as heterosexual partners. Meanwhile, activists strongly believe gay partners are able to care for a child Just as well, or even better than heterosexual partners. Whereas, sociologists and psychologists question whether or not children exposed to only one gender will be raised with a balanced understanding of gender roles and personal gender identity Chittom, par. 6). Children with a single heterosexual parent face the same situation. Even so, some argue children raised in a family with parents of only one sex are more likely to become gay when older. Gay adoption puts children who are already compromised from their experience in the foster care system at even greater risk of trauma (Chittom, par. 1). Pearson maintains there is nothing to support that children being raised in a loving family are harmed in any way by their parents sexual orientation. Pearson believes no child should be denied a home Just because of the exual orientation of the parents wanting to adopt.

Group Communication Essay Effective group communications come in forms of verbal and non-verbal techniques. Essential parts of the entire groupââ¬â¢s contribution are that the group contains full participating members, the group is diverse, and that the diversity is recognized and respected (Hartley, 1997). In the videos viewed, three were evaluated on the effective and ineffective communication skills of the participants and suggestions made on how they could improve. The videos are titled, ââ¬Å"Planning a Playgroundâ⬠, ââ¬Å"Helping Annieâ⬠, and The Politics of Sociology. Planning a Playground For the impact diversity plays on group memberââ¬â¢s communication styles, I choose the video titled ââ¬Å"Planning a Playgroundâ⬠. In this video, community residents are meeting for the first time to discuss an issue pertaining to funding of new neighborhood playground. The residents belong to diversified backgrounds in regards to their ages, genders, race, and SEC (Planning a Playground, 2008). The origins of the residents differ as one member is Black; one is Asian, whereas the other three are Caucasian. The residents come from different SEC backgrounds. One member opens the discussion about funding and how much difficulty they are having in raising enough money. Another member chimes in and mentions how she is close friends with a person with a lot of resources who might help with contributions. The group members communication style is totally different because their diversified backgrounds. Some sit facing the otherââ¬â¢s and makes constant eye contact. Another sits at the ââ¬Ëheadââ¬â¢ of the table able to view everyone but does not necessary look at everyone. Some members are taking detailed written notes, where another is just sitting there listening to everyone. Physical placement places a huge effect and impact on communication. People facing each other, sitting in close proximity, ensures everyone is heard and gives a feeling of comfort. Other effective communication methods could have made use of for the meeting and benefitted all members. Two of these methods are written materials and telephones (Hartley, 1997). Written material could be repared before the group met and used to make occurrences clearer to members, who have no knowledge of the subject. The other method is telephones, which can assist the members to gain better understanding of each other. In the use of telephones there is the reduce chance of discrimination since the members can not see each other (if they have not met prior- in some incidences here). Annie To address the verbal and nonverbal interaction among the members of a group the video titled ââ¬Å"Annieâ⬠best illustrates this. In the video there is clearly both listening and miscommunication going on. The setting of this video is that the title character, Annie is a high school girl, who seems to be suffering from a possible eating disorder as well as depression (Understanding Relationship- Helping Annie, 2008). A nurse from a school has called upon a meeting with a psychiatrist and a social worker to seek out a possible treatment plan for Annie. In ââ¬Å"Annieâ⬠, again the physical setting and placement of participants play an important part in the communication style. Two of the members are sitting side-by-side on a couch while the third person sits in her desk chair across from the couch. The person in the chair is not only able to make eye contact with the other two but also is in a physical position to see their verbal and non-verbal communications. The two on the couch are at a disadvantage for reading the non-verbal language and making eye contact with each other. The school nurse, psychiatrist, and social workers are engaged in a verbal conversation. The psychiatrist is talking to the school nurse and is not all concerned or ready to take any sort of arguments from the social worker. He is engaging in various sorts of non- verbal communication with the social workers showing his disinterest. The various non- verbal communications used by the psychiatrist are body language and voice. He continues to cut the social worker off in mid-sentence, not allowing her to speak (Understanding Relationship- Helping Annie, 2008). These non- verbal communications were hindering the process of group communication. The two methods of communications, which could have facilitated the group would have been one-to-one conversation as well as written communication. Each member could have met individually to avoid interruptions and bad sitting placement (Hartley, 1997). Also since each one evaluated Annie separately the could have just submitted their findings and suggestions/recommendations to the social worker and let her develop a plan off of that- and send a copy to the nurse and psychiatrist for editing and review. The Politics of Sociology The third video, The Politics of Sociologyâ⬠, demonstrates good communication techniques from all members of the group. The various listening techniques, which have been used by the members of this group, are encouraging, paraphrasing, and reflecting feelings. The encouraging technique used, refers to the utilization of a variety of verbal and nonverbal to promote others to go ahead with their talks. Paraphrasing is restating the conversation to give an impression that the person listening has understood the conversation. Reflecting is concerned with showing empathy with the speaker. Listening is an important activity of group communication. To bring out an effective solution or conclusion to the matter discussed, it is important to listen to all the opinions. The participants are all professors at a university but still have different backgrounds (The Politics of Sociology, 2008). There are differences in the ages, races, and subjects they teach. Each person has a different communication style. Some shot ideas out right away where others waited for all to speak then evaluate and give their opinions, while staying respectful and understanding of each otherââ¬â¢s point of view. The physical set-up of the room plays a positive part in their effective communication. Each member is facing each other and is able to see the faces of everyone. This positioning helps cut back on misunderstandings or non-verbal cues. Even though the communication techniques used in this video seem to be effective, there are always room for change and improvement. Since the group was composed of multiple people, five or six, a visual display could have been used. Either a blackboard or overhead projector could have been set up and allow each member to put up their pros and cons for change of the curriculum as well as any ideas or suggestions they had. After each member went then one person could be delegated to summarize the lists and come up with final recommendations to be voted on (Hartley, 1997). Another method could have been to break the group up into two smaller groups to work on solutions and then bring them back to the large group for discussion. Conclusion In problem solving, group discussion, or just typical dialogue it is important to have effect communication techniques. Depending on the group or topic of conversation there are several techniques that work better for discussion then others. Taking into consideration time frame or group size written, telephone, or one-on-one techniques can be utilized. Communication styles vary among groups but key essentials are universal; listening, respecting, and appropriate interaction between members.

à ±-glucosidase Inhibitory Effect of Coffee Abstract The activity-based fractionation of coffee solutions by a series of chromatography techniques led to the isolation of an active compound I which exhibited a strong inhibitory activity against à ±-glucosidase. The structure of compound I was established as norharman (9H-pyrido[3.4-b]indole) on the basis of HR-FAB-MS, 1H NMR, 13C NMR and 1H-1H COSY spectra. Compound I potently inhibited à ±-glucosidase in a concentration dependent manner but it did not exhibit any significant activity against other glycosidases. A Lineweaver-Burk plot revealed that its inhibition mode of enzyme was uncompetitive with a Ki value of 0.13 mM. Keywords: à ±-glucosidase inhibitor, à ²-carboline, norharman, coffee, uncompetitive inhibitor Introduction Coffee is the most commonly consumed beverages in the world and the health benefits of coffee consumption have been extensively studied [10]: coffee has strong antioxidant properties in vivo [16, 18] and also reduces the risk of Parkinsonââ¬â¢s [11] and Alzheimerââ¬â¢s diseases [4]. Recent studies have demonstrated that habitual coffee consumption is related to a significantly lower risk of type 2 diabetes [17, 19], but it remains unclear what mechanisms and what coffee constituents are responsible for the observed association. Animal and in vitro studies have suggested several plausible mechanisms for a beneficial effect of coffee on glucose metabolism: increase in insulin sensitivity [14], inhibition of glucose 6-phosphatase [2], an increase of glucagon-like peptide I concentration [15], and decreases the rate of intestinal absorption of glucose [12]. The à ±-glucosidase is essential for carbohydrate digestion because carbohydrates must be degraded enzymatically in the intestine before they can be absorbed. The inhibition of à ±-glucosidase slows down the process of dietary carbohydrates digestion and avoids postprandial hyperglycemia that plays a central role in the development of chronic diabetes associated complication [8]. Thus, à ±-glucosidase inhibitors have exhibited high promise as therapeutic agents for the treatment of metabolic disorders, such as type II non insulin dependent diabetes, obesity, and hyperglycemia [3]. This work was intended to evaluate à ±-glucosidase inhibitory effect of coffee previously reported as hypoglycemic and characterize the active principle isolated from coffee. Materials and Methods General p-Nitrophenyl (PNP)-à ±-D-glucopyranoside, PNP-à ±-D-mannopyranoside, PNP-à ²-D-glucopyranoside and PNP-à ²-D-galactopyranoside were purchased from Sigma-Aldrich (St. Louis, MO, USA). Yeast à ±-glucosidase, almond à ²-glucosidase, E. coli à ²-galactosidase, jack beans à ±-mannosidase, rat intestinal acetone powders, and norharman were also obtained from Sigma-Aldrich. Unless stated otherwise, all further chemicals were purchased from Sigma-Aldrich. All the reagents were of analytical grade. The UV spectrum was recorded on a Shimadzu model UV-160 spectrophotometer. High- resolution FAB mass spectra were obtained with a JEOL model JMS-AX505 HA spectrometer. 1H-NMR and 13C-NMR spectra were obtained on a Brucker AV 500 spectrometer operating at 500 and 125 MHz, respectively. (CD3)2CO was used as the solvent. Enzyme inhibition assay The intestinal à ±-glucosidase inhibitory activity was determined as described previously with a slight modification [5]. The rat intestinal acetone powder was suspended in 100 mM sodium phosphate buffer (pH 7.0) and centrifuged at 12,000 rpm for 15 min. The resultant supernatant was used as the source of the small intestinal à ±-glucosidases. For the assay of inhibitory activities of maltase and sucrase, the reaction mixture consisted of crude enzyme solution, 20 mM maltose or 200 mM sucrose, 100 mM sodium phosphate buffer (pH 7.0) and a given amount of inhibitor (50% dimethyl sulfoxide solution) in a total volume of 0.5 ml. After the reaction mixture was incubated for 15 min at 37 à ¢Ã¢â¬Å¾Ãâ, reaction was stopped by heating the mixture at 100 à ¢Ã¢â¬Å¾Ãâ for 5 min. The à ±-glucosidase activity was estimated by measuring the liberated glucose amount using the glucose oxidase method. Prior to measuring the glucose amount, the interfering agent, phenolic compounds were r emoved from reaction mixture by passing through a basic alumina column (1 x 3 cm). Acarbose was used as the positive control. The enzymatic activities of the various glycosidases were determined spectrophotometrically by monitoring the release of p-nitrophenol from the appropriate p-nitrophenol glycoside substrate [13]. The assay solutions and the potential inhibitors were added to a 96-well plate as follows: 20 L of 0.1 M phosphate buffer (pH 7.0), 20 L inhibitor, 10 L enzyme (1 U/mL), 10 L of 25 mM substrate and 40 L of methanol. Following incubation at 37 à °C for 15 min, the assay solution was stopped by adding 300 L of 1 N NH4OH solution. The glycosidase activity was determined by measuring the amount of 4-nitrophenol released from p-nitrophenol glycoside substrate was determined with a microplate reader model 550 (Bio-Rad, CA, USA) at 405 nm. All of the analyses were performed in triplicate. The concentration of the inhibitor required for inhibiting 50 % of à ±-glucosidase activity (IC50) was calculated by adjusting the experimental data (% inhibition versus the concentration of the inhibitor) to non-linear regression curves. The mechanism of enzyme inhibition was assessed by analyzing the double- reciprocal Lineweaver-Burk plot. Isolation of inhibitory compound from coffee Filtered brewed coffee was prepared in a household coffee maker: 75 g of ground roast coffee of Columbian Supremo (Arabica variety) and 500 ml water to give a brewed coffee. Commercial instant coffee (Tastersââ¬â¢ choice, Nestle) was made by dissolving 75 g instant coffee in 300 ml of hot water. Filtered brewed coffee and instant coffee solutions were separately centrifuged at 12,000 rpm and room temperature for 15 min, and used for isolation of à ±-glucosidase inhibitor. The supernatant was adjusted to pH 9 with 1 N NaOH and extracted with ethyl acetate. The ethyl acetate layer was then extracted with 0.1 N HCl solution. This acidic solution was again adjusted to pH 10 with aqueous ammonia and extracted with ethyl acetate. The organic layer containing basic components was subsequently evaporated in vacuo. Forty batches of the above ethyl acetate extracts (total 3 kg each of ground coffee and instant coffee) were concentrated and subjected to silica gel column chromatography with an isocratic solvent system of chloroform-acetone (70:30). Fractions containing the active compound (F3-F6) were combined, evaporated, and subjected to a Sephadex LH-20 column (3 x 35cm) with MeOH as an eluent. Fraction number 10-12, which showed a high inhibition and a similar TLC profile (silica gel 60 F254, Merck, chloroform:acetone = 1:1, rf 0.2) were combined and further purified. The final purification of the active compound was achieved through semi-preparative HPLC separation on a reversed phase C18 column (à ¼Bondapak, Waters, Milford, MA, USA) eluting with 75 % MeOH and detected through absorption at 254 nm. The retention time was 14.5 min. After removing the HPLC solvent in rotary evaporator, the active compound was obtained as a white powder by crystallization from cold acetone. Results and Discussion Both instant coffee and ground brewed coffee solutions inhibit à ±-glucosidase enzyme activity. Instant coffee showed a slightly higher degree of inhibition than brewed coffee (Data not shown). The activity-based fractionation of coffee solutions by a series of chromatography techniques led to the isolation of an active compound I (2.24 à ¼g/ g of roasted ground coffee; 3.85 à ¼g/ g of instant coffee) which exhibited a strong inhibitory activity against à ±-glucosidase. The isolated compound I was shown to be chromatographically pure by TLC and HPLC with various solvent systems and deduced to be a nitrogen-containing compound based on a positive reaction to Dragendorffââ¬â¢s reagent. The UV spectrum of the compound in methanol exhibited absorption maxima at 230, 285 and 348 nm. The molecular formula of compound I was determined to be C11H8N2 (M+ m/z 168.0736; calcd. 168.0688) by high resolution mass analysis. 1H NMR spectrum of compound I showed 7 aromatic proton signals (à ´7.2-8.9 ppm) and one free proton signal (à ´10.63 ppm). 13C NMR spectrum showed 11 carbon signals around 110-145 ppm (Table 1). Taken together, the structure of compound I was deduced as à ²-carboline, norharman (9H-pyrido[3.4-b]indole, Fig. 1) with 1H NMR, 13C NMR, and 1H-1H COSY spectra and confirmed by comparison of physical data with those of the authentic specimen. Compound I potently inhibited à ±-glucosidase in a concentration dependent manner, but it did not display any significant inhibitory effects against à ²-glucosidase, à ±-mannosidase, and à ²-galactosidase when tested at a concentration of 10 mM (Table 2). The inhibitory profile demonstrated that the activity of compound I was greater against maltase compared with sucrase (IC50 values: 0.27 mM for maltase and 0.41 mM for sucrase). Although the inhibitory potency was weaker than that of therapeutic drug acarbose (IC50 value: 0.18 mM for maltase and 0.02 mM for sucrase), observed data clearly indicated the potential of compound I as an à ±- glucosidase inhibitor. The pre-incubation of compound I with the enzyme increased the inhibition of à ±-glucosidase activity, implying that this compound reacted with the enzyme slowly. The à ±-glucosidase activity was fully restored when the enzyme was incubated with an amount of compound I which could inhibit enzyme activity up to 90 % follo wed by eliminating the compound I with a PD 10 desalting column (Pharmacia, Piscataway, NJ, U.S.A). This result demonstrated that compound I was a reversible inhibitor. A double-reciprocal Lineweaver-Burk plotting under various amounts of compound I showed linear lines intercepting on 1/V axis in parallel. The kinetic data suggested that the compound I was an uncompetitive inhibitor, with a Ki value of 0.013 mM (Fig. 2). As a result, compound I, a reversible uncompetitive inhibitor of à ±-glucosidase, was isolated from coffee and identified as an active principle. When compound I was given in combination with a carbohydrate-rich diet orally, the postprandial plasma glucose levels were significantly dropped in non-diabetic rats (unpublished data). Compound I, a tricyclic indole à ²-carboline alkaloid norharman is distributed widely in biological systems and exhibits a wide spectrum of pharmacological and neurological effects: antidepressant and antianxiety effects in rats [7], inhibitory activities of monoamine oxidase and nitric oxide synthase [9], as well as an increase of insulin secretion two- to threefold from isolated human islets of Langerhans [6]. However, à ±- glucosidase inhibitory activity of norharman has not previously reported. Coffee has been noted as the primary exogenous source of norharman. A high variability in à ²-carboline content of coffee samples was observed between coffee species (arabica, robusta) and also depended on roast degree and instant coffee production process. An average of 3 cups of coffee per person per day could account for an ingestion of up to 72 à ¼g of norharman [1], although this will depend on the coffee strength. Coffee contains numerous substances. However, little is known regarding the effects of individual constituents on glucose metabolism. The cohort study has supported that the most prominent coffee compound caffeine is irrelevant to risk of type 2 diabetes [20]. Chlorogenic acid, the most abundant polyphenol in coffee, has been shown to reduce glucose concentrations in rats, caused by increasing insulin sensitivity as well as reducing hepatic glucose output through inhibition of glucose 6-phosphatase [12]. Without excluding any other possible mechanism, this report observes à ±-glucosidase inhibitory activity as a possible mechanism of hypoglycemic effect of coffee and assigns à ²-carboline alkaloid norharman as one of active principles in coffee. Coffee appears to contain active principles other than norharman as evidenced by several active peaks in chromatography systems. It may be possible that various active constituents in coffee act synergistically against à ±-glucosidase acti vity. Characterization of other active principles is under progress. Capital Punishment Introduction: Think for a moment about the fall into sin. Think about the people in Noahââ¬â¢s day. Sodom and Gomorrah. What is the punishment inflicted upon all of them by God? Death. The death penalty, or capital punishment, is the first and harshest penalty for sin. Although it has evolved and changed form over the years, and is now decided upon by men playing God, it is still an acceptable, God pleasing form of punishment. I am going to tell you what capital punishment is, its history, its current role in the justice of criminals, and why I believe that it pleases God. Body: Capital punishment, simply put, is killing someone for a crime that they have committed. This crime is murder. In some countries and states, it is considered acceptable to kill the person who has taken the life of another. The methods of execution have varied since the beginning of time, but three current popular ways include the gas chamber, lethal injection, and the electric chair. My overhead shows the different ways that states used for execution in 1994. In addition to the three most popular choices, a hanging and a firing squad are also used in a few states. According to the book ââ¬Å"Should We Have Capital Punishmentâ⬠by JoAnn Bren Guernsey, these choices were made with the goal of a more humane murder in mind. The gas chamber is a small, sealed room in which the prisoner sits strapped to a chair. A lethal gas is sent through the floor of the room, and death usually takes about five minutes. Lethal injections simply involve the insertion of a needle filled with poison into a vein and injected. This procedure can be effective, but also takes long amounts of time quite often. The electric chair was invented as a way to quickly and painlessly kill the prisoner, but has proven to not be as effective as thought. It has taken up to three 2000 volt shocks to kill someone, with fire, sparks, and smoke reported to be seen coming from their head. None the less , the electric chair is currently the most popular method of execution. The history of capital punishment goes back to the beginning of time. The punishment for the fall into sin was death. God first punished Adam and Eve by banishing them from the Garden of Eden, and forcing them to endure pain and conflict as a reminder of their sin.

Allowing the community to become involved in the project will make them feel invested in their environment and will help produce an environmentally healthy awareness amongst the islanders. The first two factors obviously favor the Caribbean Island in that it is located closer to the equator and to sea level. New York is at a higher elevation and is not close to being in any tropical zones. In terms of isolation, neither location seems to be at an advantage over the other one. The New York habitat is located in an industrial sector meaning that it is not connected with extensive forests that could contribute to the diversity of the area. Islands obviously experience a notable amount of isolation but this does nothing to cancel the effect of its latitude location. The industrial sector undoubtedly leaks a significant amount of chemical pollution into the area that may hinder the progress of achieving successful reforestation. An analysis of the setting in Nathaniel Hawthorne's "Young Goodman Brown" In the story of "Young Goodman Brown" setting plays an important role. It provides symbolism to certain events and provokes emotions amongst the characters, especially those of Goodman Brown. The story of "Young Goodman Brown" is that of a man on an adventure to feed his curiosity and to visit the dark side of his Puritan town. Once he arrives at the destination of his adventure, he realizes that many of his elders have followed in the paths of evil and that holiness and innocence has been vanquished from his once thought to be holy Puritan town. The central idea of "Young Goodman Brown," is the conflict in Goodman Brown between joining the devil and remaining "good." It is a very difficult journey for Brown, as he travels through the woods, all the while thinking of the "good" things (like his wife Faith) he would be leaving behind. This internal conflict ultimately destroys the Young Goodman Brown and creates a new man. At the beginning of the story Goodman Brown sets out on his journey at sunset; to set out at sunset is symbolizing darkness, which in turn symbolizes evil, which begins the setting for the story of "Young Goodman Brown." As Brown is leaving he kisses his wife, Faith, goodbye; the name Faith is intentionally used to symbolize the faith in god that they both share and also what Brown is leaving behind to go on his journey. Brown's wife, Faith, is also wearing pink ribbons in her hair, which give the impression that she is an innocent godly woman. Right before Brown leaves on his journey Faith says, "pr'y thee, put off your journey until sunrise." (309). This is showing that sunrise is a more pleasant and peaceful time to go on a jou... ...ed by the devil tempting Brown to join the dark side. The way in which the devil lures Brown is by convincing him that his relatives and town's people have already crossed over to the dark side and denied their faith. The struggle that Brown is now facing is whether to follow is relatives and town's people or to follow his faith in god and deny the devil, that is Brown's internal conflict. These internal and external conflicts are what turned Brown into a dynamic character by the end of the story. Instead of believing and trusting that his relatives and friends are godly, he now knows that they have all been tempted by the devil and many have crossed over to the dark side; he went from being trusting to skeptical of his friends and his faith. Certain advantages of living in a large city City life and rural life are absolutely the different sides of the coin, so choosing whether to live in a big city or a small town requires serious consideration. Each city has its unique aspects and qualities that distinguish itself from others. Each person has his or her own opinion about where to live based on his or her life style. However, according to me, living in a large city has certain advantages over living in a small town: convenient public transportation, greater entertainment facility and higher employment. To begin with, a metropolitan city has its obvious advantage when it comes to the public transportation. It is wildly known that a big city has extremely convenient transportation which can be best illustrated by the example of New York, where the subway runs twenty-four hours for seven days a week and there are tremendous cabs and buses everywhere. Not to mention that the transport system is always fast, safe, clean and a ffordable. For the poor, who can only rely on the public transportation, such a convenient and efficient system makes it much easier for them to seek work opportunities far from their living houses and also gives them greater access to education, healthcare and recreation. In addition to public transportation, what makes people pour into big city is its wonderful entertainment facility. Many small towns often offer less social amenities than expected due to its demographic and geographic factors. Whereas, countless facilities of entertainment and cultural activities such as cinemas, theatres, concerts, museums can be found in a large city like Shanghai with the least effort and time. Such facilities not only offer people a place where they can be relaxed after a weekââ¬â¢s hard work, but also can nurture the soul and cultivate the mind to some extent, which is particularly beneficial to children who are willing to visit some historical museums during their spare time. Finally, higher employment is another reason why people prefer urban areas. On the one hand, there are a lot of headquarters for large companies in major cities as well as large factories, so the average employed person is a step higher on the ladder than they will be in smaller cities. And it is likely that people will find something that specifically matches their skills. For instance, a friend of mine who majors in software engineering can easily find a related job in big cities while he may find the different line of work if he is in rural areas. On the otherà hand, statistics show that people in large cities are more likely to have a Bachelorââ¬â¢s degree or better, which contributes to the high employment and better payment. Taken all of the above reasons into consideration, small towns cannot be compared with big cities in public transportation, entertainment facilities and job opportunities. The team that scores the highest points will be rewarded with a prize, such as vouchers, etc. 4. 2 Links to survey results The action plan created was based on a census of the whole school to observe whether they would participate in athletics and their reasons to not. To justify the particular choices created in the action plan by showing the statistics which have guided the development as the spikes in the statistics assist in improving the participation by surveying the trend. The reason a record chart was implemented as it adds a competitive flair and students receive social rewards within the athletics carnival as 19% of students feel that there is no reward for students if they win the events, this will help students strive for the record instead of just trying to win. A massive 27% of students feel that they are not good enough for the athletics carnival and believe there is no point to participate and simply just socialize with their peers, with the appearance of a famous athlete; students would be motivated to part take in the events as the special guest can provide moral support and advice to improve their technique, etc. during the carnival as they can join in with the students. By creating team based events the 80% of students that prefer team sports are able to participate and at the same time fill the social void according to the 25% as they strive to motivate fellow teammates and allow each other perform better overall, also considering the 66% that would participate in the carnival if their peers were to join in. Biblical Worldview Essay Essay
We all have a set of values and beliefs that help shape the way we view the world. While many of us can share the same tools that shape our worldview; everyoneââ¬â¢s is unique to themselves. Romans 1-8 provides valuable teaching regarding the natural world, human identity, human relationships, and culture. These 4 categories make up the majority of the world and life as we know it. As Christians, this passage in the Bible should hold a great influence on our worldview. I will outline what Romans 1-8 teaches us about each category and how that affects my personal view of the world. As Christians, we know that God created the universe and everything in it by simply speaking. Everything in His creation attests to Him and His glory. This allows people who may never actually hear about Christ to still be able to see Him through creation. There is a verse in Luke 19:40 that goes on to say that even the rocks will cry out if everyone is silent. Romans 1:20 says, ââ¬Å"For since the crea tion of the world Godââ¬â¢s invisible qualitiesââ¬âhis eternal power and divine natureââ¬âhave been clearly seen, being understood from what has been made, so that people are without excuse.â⬠Through the natural world, God has provided a way to show Himself to all mankind. When I am out in nature on a beautiful quiet day I can feel the Lordââ¬â¢s presence. When I am on a ship in the middle of the Atlantic Ocean watching the sun set, I can see the Lord. I am young and have not yet seen many of the natural wonders of this world. I have seen enough though to know that God is present throughout nature. I donââ¬â¢t understand how a person could look out on a beautiful scene of nature and not feel God. He truly does speak through the beauty of His creation. Our human identity is who we believe ourselves to be. Without God I know that I am a destitute and wicked creature. We are capable unspeakable evils. I have hope though through my redemption. I know that any good thing I do or obtain is not of myself but from God. God created us in His image so we can catch a glimpse of what He is truly like. However, as humans we are wicked and evil in heart. Nothing but the love of C hrist can save us. Romans 3:10-18 clearly lines just how unrighteous and devoid of hope we are within ourselves. This passage shows us our true human identity without Christ. We have hope though because we have been given Christ! Romans 8:26 tells us that Christ intercedes for us at the feet of God. Nothing can separate us from the love of Christ once we accept Him!à (Romans 8:35) Relationships make up a huge part of our lives. The Bible has a lot to say about relationships and how to treat one another. Relationships make an enormous impact on our world view because we are influenced by the people in our lives. Romans 1:28-32 lists the evils things that we do to one another such as envy, murder, strife, deceit, malice, and gossip. This is just a fraction of the sins that cause us to have damaged relationships with each other and ultimately God. How could you come before God and expect a good relationship if you have a broken relationship with someone in your life? Jesus teaches us to love one another and forgive over and over. When your heart is hardened towards another person, then you are u ltimately hurting your own relationship with God as well. The second chapter in Romans cautions us against judging others. We have to treat one another the way we would want to be treated; with kindness and mercy. We also have to be careful about what kind of people we have relationships with. If you surround yourself with evil people then you will be influenced to do evil. During the time that Paul was writing Romans, there was a religious hierarchy in place. The Pharisees were the leaders and aristocrats in the Jewish community. They were extremely strict and followed a huge list of rules. They were so caught up in their rules that they couldnââ¬â¢t open their hearts and minds to Jesus. Also at this time it was widely believed that only the Jewish community could be saved through following the rules and the Gentile population was doomed. Romans 3:23-25 teaches us that, ââ¬Å"for all have sinned and fall short of the glory of God, and all are justified freely by his grace through the redemption that came by Christ Jesus.â⬠Jesus death paved the way for everyone to be saved, not just the Jews! We learn that we are saved through faith and not by works. We can never earn our salvation. Todayââ¬â¢s culture in America is so focused on material possessions and gaining more. We are caught up in our shiny toys and items much like the Pharisees were caught up in their rules. We have to stop getting caught up in things that donââ¬â¢t really matter and see what God is trying to use us for. We all have a purpose and a reason for being here. If you are not living your life to further Godââ¬â¢s message than you are wasting it. Our culture today makes it so easy to forget what we are really supposed to be living for. As Christians we must be vigilant and not let the snares of this world snag our heart and divert it from our true calling. Everyone has their own uniqueà view of the world and reasoning for their views. Christians build their world view with the structure that the Bible has provided us. Paul writes about the natural world, our identity as people, how to treat others, and about the culture of the world. We are not perfect and can never hope to be but the grace of Jesus offers us salvation. Romans 8:12 reminds us that we have an obligation to live a life through the Spirit and not by the flesh. Every day we must work harder and strive for righteousness and a Christ-like attitude. Our worldview affects the way we live and can have an impact on all those around us. The better our worldview, the better our light will shine for Christ.

An Ankle Sprain of a Female Colligate Basketball Player Objective: To introduce a case study about a mild ankle sprain. Background: The most common way to sprain an ankle in basketball is to step on someone elseââ¬â¢s foot, or to plant and to turn the wrong way. Differential Diagnosis: Could be an injury to the deltoid ligament, CF, or the ATF ligament. Treatment: Ice, High volt, Theraband, Towel scrunches, Towel stretches, Whirlpool Conclusion: Rehab will help her get her strength back in her ankle as long she continues to work hard. Keywords: Ankle sprain, High volt, Medial and Lateral Malleolus Objective The ankle joint is the most commonly injured part of the lower leg. It happens from an unusual twisting action when the foot is planted awkwardly or when running on uneven ground. An unbearable amount of force is placed on the joint itself. Such injuries occur all the time in athletics or just by running. The ankle is made up of bones, tendons, and ligaments. The major bone of the lower leg is the tibia; it holds most of the bodyââ¬â¢s weight. It is made up part of the medial malleolus, which consist of the inside up hump of the ankle. The fibula is the next largest of the ankle bone in the lower leg. It forms the lower leg end form the lateral malleolus, the outer hump of the ankle. The smallest ankle bone is the talus which completely makes up the bone on the top of the foot. The tendons connect the muscle to the bones. There are several muscles that help control motion at the ankle. The tendon connects one or more of the bones to the foot. Tendons can be stretch to torn when a great amount of tension is placed upon it. They can also be pulling away from the bone, such as the Achilles tendon rupture. Ligaments provide connections between the bones. Ligaments are mostly sprained. The ankle has many bones that comes together to form the joint. The most commonly injured ligament is the anterior talofibular ligament that connects the front of the fibula to the talus bone on the front outer rim of the ankle joint. Ligaments are sprained when a great than normal force is placed on it. This is done when the foot is inverted most of the time. This happens when the foot is awkwardly planted or is stepped upon during activities. Stepping in a surface that is irregular, such as in an athletic event when one player steps on another player foot a sprain can result. Background A twenty-one year old female, basketball player experienced an ankle sprain by accidentally stepping on another playerââ¬â¢s foot. The player was going up to make a shot landed on the opponents foot when she came back down, which made her ankle invert. The head athletic trainer evaluated her then taped her ankle to provide support and keep the swelling to a minimum at that time, so the player could return to play. Immediately after the game, the playerââ¬â¢s ankle was iced down to control the swelling and was receiving NSAIDââ¬â¢s to help with the pain or discomfort she was feeling. The player was referred to the team physician for x-rays and MRI to help rule out fractures. Treatment was started to help relieving the swelling and pain. Treatment In the first couple of days of rehab, she received high volt, and ice to help with the swelling she also did ankle pumps that also help with moving out the inflammation. As each day went by, towel scrunches, towel stretches were performed as three set each. Compression pumps were administered for 15 minutes several times a week, rhythmic stabilization, and aquatic therapeutic exercises were added as she got closer to the sub acute phase. The sub acute phases are about six days to until six weeks. In this stage she was allowed to jog straight ahead, trying wide figure-eight at first. Eventually the player was either on the bike or the stair stepper for 10 minutes a day. After the bike she was moved to the shuttle press with four cords, and three sets of ten, after the shuttle she did calf raises. The farther she progressed, the whirlpool was introduced, and both hot and cold water. Two set of tens, in all four directions using Therband was the next step of treatment; ultrasound was done to help break up scar tissue that had started to form. As her ROM improved along with other tests, she was moving closer to the return to play phase. The return to play phase this is when functional test and sport specific drill can be started. Functional testing is important along with continuing with other exercises and modalities. She moved to three set of fifteen with a Therband in all direction, hot pack assisted with high volt was added. Following the Therband the shuttle with the dynadisc, doing three set of fifteen along with four cords, she also did towel scrunches five times; wobble board, marbles and exercise. ROM exercise was done to increase plantar flexion and dorsiflexion. Differential Diagnosis There are many kinds of ankle injuries that can be present in variety of ways. The calcaneocuboid joint injury is a kind of inversion ankle sprain that involves the ligament overlying that joint. Which cause immediate swelling, pain and tenderness to the touch? The pain is localized to that region of the joint. The deltoid ligament resists abduction and lateral rotation of the ankle. However sudden forceful motion of the ankle may tear the ligament or stress it. The ligament may avulse that is attacked to the malleolus; a vast majority of case, there will be a tear through the ligament. The tear could be associated with compression of the talar joint. There could be an injury also to the ATFL and the CF ligament. Conclusion The player went through all the appropriate phases to get back in to the return to play phase. She will continue to do rehab so that her ankle with improve and get stronger.
